摘要
目的探讨HLA-Ⅰ类、-DQA1、-DQB1等位基因与汉族人群斑秃相关性。方法采用聚合酶链反应-序列特异性引物(PCR-SSP)方法,检测192例汉族斑秃患者和252例正常对照的HLA等位基因频率。结果①斑秃患者HLA-A*02、-A*03、-B*18、-B*27、-B*52、-Cw*0704、-DQA1*0104、-DQB1*0604和-DQB1*0606等位基因频率较对照组显著性升高;②某些等位基因与斑秃早发和发病持续时间长有关。结论HLA-A*02、-A*03、-B*18、-B*27、B*52、-Cw*0704、-DQA1*0104、-DQB1*0604和-DQB1*0606等位基因可能是斑秃易感基因或与易感基因相连锁;不同发病年龄和发病持续时间的斑秃患者在遗传背景上可能存在异质性。
Objective To perform genotyping of HLA class I, -DQA1 and -DQB1 alleles in order to identify HLA association with alopecia areata (AA) in Chinese Hans. Methods polymerase chain reaction-sequence-specific primer (PCR-SSP) method was used to analyze the distribution of HLA- class Ⅰ, -DQA1 and -DQB1 alleles among 192 patients with AA and 273 healthy controls. Results (1) The frequencies of HLA-A * 02, -A * 03, -B * 18, -B * 27, B * 52, -Cw * 0704, -DQA1 * 0104, -DQB1 * 0604 and -DQB1 * 0606 alleles were significantly increased in patients with AA compared with the controls. (2) HLA-DQB1 , 0606 was observed significantly increased frequency in early onset patients compared with the controls, so were HLA-A * 03, -B * 27 and DQB1 * 0604 in late onset patients. (3) The frequency of HLA-DQA1 *0104 and -DQB1 * 0604 alleles were significantly increased in patients with long duration compared with controls. Conclusion HLA-A * 02, -A * 03, -B * 18, -B * 27, B * 52, -Cw * 0704, -DQA1 * 0104, -DQB1 * 0604 and -DQB1 * 0606 alleles may be AA susceptible alleles or may be in close linkage with the susceptibility genes. There may be different genetic backgrounds in AA patients with different age of onset and duration.
