摘要
目的检测鼻腔 NK/T(natural killer)细胞淋巴瘤中 EB(Epstein-Bart)病毒感染潜伏膜蛋白(latentmembrane protein-1,LMP-1)与细胞因子κB(nuclear foctor-κB,NF-κB)的表达及细胞凋亡指数(apoptotic idex,AI),研究 EB 病毒与鼻腔 NK/T 细胞淋巴瘤的关系。方法 26例鼻腔 NK/T 细胞淋巴瘤为实验组,30例良性淋巴组织反应性增生病例为对照组。应用免疫组化 SP 法检测石蜡包埋块中 LMP-1和 NF-κB蛋白的阳性率,DNA 末端标记技术(TUNEL)检测 AI,比较这些指标在实验组和对照组中的差异,分析三者间的相互关系。结果 LMP-1检出率、NF-κB蛋白表达阳性率、AI 在实验组分别为69.2%(18/26)、65.4%(17/26)、2.31±0.38;对照组分别为10%(3/30)、30%(9/30)、3.12±0.45。两组指标间差异均有统计学意义(P<0.01)。LMP-1检出率与 NF-κB蛋白表达阳性率呈正相关(P=0.0010,r=0.6860),与细胞凋亡指数呈负相关(P=0.0001);NF-κB蛋白的表达与细胞凋亡率呈负相关。结论 EB 病毒感染与鼻腔 NK/T 细胞淋巴瘤的发生及发展关系密切。(中国眼耳鼻喉科杂志,2007,7:31~32)
Purpose To examine expression of latent membrane protein-1 (LMP-1), nuclear factor κ gene binding (NF-κB) in nasal NK/T cell lymphoma, and to explore their relationships to apoptosis.Methods Twenty six cases of nasal NK/T cell lymphoma(test group) and 30 cases of benign lymph follicle hyperplasia (control group) were collected in this study. Expression of LMP-1 and NF-κB were examined with immunohistochemical technique in the tissue specimens dissected from the lymphoma or hyperplasia and apoptotic idex was examined by TdT-mediated dDUTP-FITC nick end-labelling (TUNEL) method in all the cases. Results In the test group the positive rates of LMP-I and NF-κB were 69.2% (18/26) and 65.4%(17/26) ,and AI, 2.31± 0.38; In the control group, the positive rasts of LMP-I ,NF-κB and AI were 10% (3/ 30) ,30% (9/30) ,3.12 ±0.45 respectively. The expression intensity of LMP-I was correlated positively to that of NF-κB ( P =0.001, r = 0.686) but to apoptotie index negatively( P = 0.0001 ). Conclusion The infection of EB virus has a close relation with the occurrence and development of nasal NK/T cell lymphoma. ( Chin J Ophthalmol and Otorhinolaryngol, 2007, 7 : 31-32)
