摘要
目的:观察高糖对体外培养的大鼠腹膜间皮细胞(PMC)细胞膜上葡萄糖转运蛋白1(GLUT1)含量的调节作用。方法:体外培养的W istar大鼠PMC置于不同糖浓度培养液中;流式细胞仪定量检测细胞膜上GLUT1的含量,生化分析仪检测PMC对葡萄糖的摄入。结果:随着细胞外葡萄糖浓度增加,细胞膜上GLUT1的表达增加,同时伴有PMC对葡萄糖的转运增加(P<0.05)。结论:葡萄糖以浓度依赖方式上调PMC细胞膜上GLUT1的蛋白表达及PMC对葡萄糖转运的增加导致PMC受损。
Objective: To study the effect of high glucose on glucose transporter-1 ( GLUT1 ) in rat peritoneal mesothelial cell (PMC) in vitro. Methods: PMC of male Wistar rats was placed in the cultures containing different glucose concentrations, The expression of GLUT1 in the plasma membrane of PMC and the glucose absorption in PMC were detected by flow cytometry and bicehemistry analyzer, respectively. Results: The expression of GLUT1 in the plasma membrane of PMC and the glucose transportation in PMC increased with the glucose concenttration( P 〈0.05). Conclusion: High glucose up-regulates the expreasioni of GLUT1 in the plasma membrane of PMC in a dose-dependent manner and increases the glucose absorption in PMC.
