糖尿病模型大鼠骨折后骨痂细胞增殖情况的观察 被引量:3

Osteoblast proliferation behavior in bony callus of rats with diabetes after fracture

摘要 目的测定糖尿病大鼠骨折后骨痂细胞增殖核相关抗原Ki67指数,观察糖尿病大鼠骨折后成骨细胞的增殖能力。方法Wistar雄性大鼠40只,随机分为两组。实验组腹腔内注射链脲佐菌素破坏胰岛细胞诱导为糖尿病大鼠,对照组为正常组。两组大鼠均折断其右胫腓骨,然后复位外固定。于骨折后第1、2、4、6周拍x线片观察骨折愈合情况,取骨痂切片行HE染色光镜下观察,免疫组化测定Ki67指数。结果实验组第1、2、4、6周X线片上骨痂生成量骨痂内成骨细胞数量均明显少于对照组;Ki67指数第2周实验组为(18.06±3.54)%,对照组为(26.87±4.61)%,两组差异有统计学意义(P<0.01)。结论糖尿病大鼠骨折愈合过程中,骨痂中的成骨细胞增殖能力下降是导致糖尿病骨折大鼠愈合差的原因之一。 Objective To detect nuclear proliferation-associated antigen (Ki67 index) in bony callus, to observe the ability of osteoblast proliferation, Methods Forty male Wistar rats were divided into two groups randomly. One was experimental group, the other was normal control group. Diabetes mellitus was induced by a enterocoelia injection of streptozotocin. Normal saline was given to control group. The right tibiofibula of all the rats were broken, and then treated with a reduction and external fixation. The bony callus formation was valued by X ray at 1,2,4,6 weeks. Bony callus sections of fracture region were treated with HE and immunohistochemical staining to detect the Ki67 index. Results The bony callus formation was less in experimental group than that in normal control group at each time point both in X ray and HE stain section, the Ki67 index at 2 week was (18.06±3.54)% in experimental group, while it was (26.87±4.61)% in control one(P〈0.01). Conclusions The inadequate osteoblast proliferation was one of the reasons for the impaired bone healing in rats with diabetes after fracture.
