摘要
目的观察糖尿病大鼠骨折前、后血清转化生长因子-β1(TGF-β1)水平变化,了解TGF-β1在糖尿病大鼠骨折愈合过程中的作用。方法雄性SD大鼠40只,腹腔内注射链脲佐菌素破坏胰岛细胞,制备糖尿病大鼠模型,同时随机分为各实验组。实验组、对照组大鼠折断右胫排骨,然后复位外固定。骨折后拍X线片观察骨折愈合情况,操作完毕分笼饲养。测不同时间两组大鼠血清TGF-β1含量。结果骨折后1、2、4周,测定实验组、对照组TGF-β1含量,两者差异有统计学意义(P<0.01),X线片上实验组骨痴延迟对照组。结论 TGF-β1在整个糖尿病大鼠骨折愈合过程中具有重要调节作用,动态观察TGF-β1含量,对判断糖尿病骨折愈合进展、骨折延迟愈合、骨不连研究提供实验数据,有利于从分子水平了解骨折愈合的调控机制。
Objective: To investigate the change of concentration of serum transforming growth factor-β1(TGF-β1) in streptozotocin-induced diabetic rats before and after fracture.To probe the effect of transforming growth factor-β1(TGF-β1) during the course of fracture healing in streptozotocin-induced diabetic rats.Methods: Totally 40 male SD rats,the rat model of diabetic was induced by intraperitoneal injection of streptozotocin.Rats were randomly divided into normal control group(NC group),diabetes mellitus group(DM group).The left tibiofibula of all the rats were broken,then treated by external fixation.The bone callus formation was evaluated by X-ray and the concentration of serum transforming growth factor-β1(TGF-β1) of the two groups was detected at different time.Results: The concentration of serum transforming growth factor-β1 of the two groups was detected at1,2,4week.The difference between them was significantly(P0.01).X-ray films showed that the fracture healing of normal control group was superior than that of diabetes mellitus group.Conclusion: The transforming growth factor-β1(TGF-β1) play an important regulation role in the course of fracture healing in diabetic rats.Dynamic observe the concentration of serum transforming growth factor-β1(TGF-β1) will provide reliable experimental data to estimate fracture healing,delayed fracture healing and nonunion.It was favourable to acquaint the regulatory mechanism of fracture healing from molecular level.
