摘要
研究了温度、pH值、溶氧对里氏木霉R3液体深层发酵产纤维素酶的影响,并进行了工艺优化.用50 L全自动通气机械搅拌发酵罐,接种量10%,发酵过程采取分段控制pH值、温度和溶氧的工艺,发酵120 h酶活力最高,FPA和CMC酶活力分别达到48.9 FPIU/mL和321.68IU/mL,FPA酶活比工艺优化前提高了221%.
The effect of temperature, pH value and soluble oxygen on the submerged fermentation of cellulase with Trichoderma reesei R3 were studied, and technics was optimized. In a 50 - litre aerate automatic fermentation tank with mechanical agitation,the dosage of inoculation was 10%, the temperature, pH value and soluble oxygen were controlled step by step. Cellulase activity of filter paper and CMC reached 48.9 FPIU/mL and 321.68 IU/mL when ferment for 120h,comparing to the original technics, cellulase activity of FPA was higher by 221%.
