部分中国野生百合亲缘关系的RAPD分析 被引量:21

摘要 采用随机引物扩增多态性DNA(RAPD)技术分析了中国野生百合25份种质的亲缘关系。从160个10bp随机引物中筛选出15个重复性好的随机引物,通过扩增共得到156个DNA片段,其中多态性谱带为133条,占85.3%。根据多态性结果进行聚类分析,将25份种质分为4类3个亚类。除野百合(秦岭种源)外,RAPD聚类结果与传统形态分类基本吻合。轮叶百合聚为第I类,轮叶组与其它组明显分开;4种喇叭花组百合聚为第II类;另4种喇叭花组百合及未鉴定的河南1号聚为第III类;第IV类含有15份种质,大部分为卷瓣组野生百合,但第IV类第3亚类含有钟花组3种野百合,表明卷瓣组和钟花组之间存在着基因交流。 RAPD analysis was used to study the genetic relationship among 25 wild lily germplasm native to China. 15 primers produced reproducible amplified bands were screened out from 160 random primers of 10-bp. A total of 156 loci were identified, of which 133 loci showed polyrnorphism, accounting for 85.3%. 25 lily germplasm could be divided into 4 clusters and 3 sub-clusters. Except for Lilium brownii (from Qinlin) , the result of RAPD of the other 24 germplasm agreed with their taxonomic classification on the whole. L. ditichum was grouped into the first cluster, and sect. martagon was remarkably distinguished from other sect. 4 germplasm from sect. martagon were grouped into the second cluster, and the other 4 from sect. martagon and Henan No. 1 were grouped into the third cluster. The forth cluster included 15 germplasm, most of which were from sect. sinomartagon; but the third sub-cluster comprised 3 germplasm from sect. lophophorurn, illustrating gene exchange between sect. sinomartagon and sect. lophpphorum.
