摘要
在百合(Lilium)杂交育种中,常采用多个父本混合花粉与一个母本杂交的方法来提高育种效率,为此,研究明确快速父本鉴定方法极为重要。采用SRAP分子标记技术对亚洲百合Red Latin为母本,亚洲百合Matrix和野生种山丹为父本混合授粉后百合杂交Fl代进行父本鉴定。引物筛选共获得4对条带清晰、多态性高的引物(Me1Em4,Me5Em9,Me7Em2和Me7Em7)用于后续SRAP分子标记分析。采用聚丙烯酰胺凝胶电泳技术,对11个F1后代单株和3个亲本材料的SRAP分子标记结果表明:用Me1Em4引物在11个F1后代单株里检测出3条父本特征条带,可以确定8个后代的父本是Matrix,2个后代的父本是山丹;用Me5Em9引物检测出2条父本特征条带,可以确定3个后代的父本是Matrix,1个后代是山丹;用Me7Em2引物检测出2条父本特征条带,可以确定5个后代的父本是Matrix,2个后代是山丹;用Me7Em7引物检测出2条父本特征条带,可以确定3个后代的父本是Matrix。综合分子标记鉴定结果,确定9个株系是Red Latin×Matrix后代,2个株系是Red Latin×山丹的后代,同时表明SRAP分子标记技术可以用于混合授粉后代父本的鉴定。
In the hybridization Breeding of Lilium, often using multiple male and a female mixed pollen hybridization methods to improve breeding efficiency, therefore, studying clearly paternal quick identification method is extremely important. Genetic relationship among 11 Flprogenies from the cross combination of Red Latin×(Matrix+Lilium pumilum DC) using a mixed-pollen cross and their parents were studied using sequence-related amplified polymorphism(SRAP) with 4 primer combinations(Me1Em 4,Me5Em9, Me7Em2, Me7Em7), in order to identify their real fathers. Our results showed that Me1 Em 4 primer pair identified 3paternal characteristic bands, indicating 8 progenies from Matrix and 2 progenies from Lilium pumilum DC; Me5Em9 primer pair identified 2 paternal characteristic bands, indicating 3 progenies from Matrix and 1 progenies from Liliumpumilum DC; Me7Em2 primer pair identified 2 paternal characteristic bands, indicating 5 progenies from Matrix and 2 progenies from Lilium pumilum DC;Me7Em7 primer pair identified 2 paternal characteristic bands, indicating 3 progenies from Matrix. From the above results, we concluded that 9 hybrids from Red Latin×Matrix were identified, and 2 from Red Latin×Lilium pumilum DC. All results obtained in this paper indicated that SRAP molecular markers were efficient in identifying paternal parent of progenies from a mixed-pollen cross.
