摘要
本实验通过添加1~4mg/ml牛输卵管蛋白至TCM-199培养液中,对小鼠原核胚进行体外培养,观察原核胚的分裂和发育情况。小鼠原核胚很少分裂,远低于对照组原核胚分裂和发育水平。牛输卵管蛋白对小鼠原核胚的发育起明显的抑制作用,同时这种抑制作用随牛输卵管蛋白浓度的降低.似乎有减轻的趋势,表明牛输卵管蛋白中,存在抑制小鼠胚胎分裂和发育的蛋白质,对小鼠原核胚的抑制作用具有剂量效应。
Bovine oviducts were Harvested fron local slaughterhouse at either the
follicular orluteal stage of the oestruse cycle of the killed cattles. Nevertheless,bovine oviducal
fluicl proteins(BOF),collected by flushing the oviduct lumen with normal saline,were attained by
lyophilizigthe dialyzed flushings. BOF used for pronuclear monse embryos culture, the
experiment was con-ducted to investigate in vitro development of one-cell mouse embryos in
TCM-199 + 25mMHepessupplemented with BOF at the concentrations of 1~4mg/ml.Cultured in
TCM-199 + BOF,cleavage to 2.cell or 3,4-cell stages hardly happened in comparison to the
control treatment.Thisstudy indicated that additlon of BOF to TCM-199 retarded embryo
development in vitro comparedto controls,But as if BOF effects on embryo Cleavage Was
dose-dependent,A reduction of thepresence of BOF in the TCM-199 resulted in embyos cleaving
( 1mg/ml BO),it was suggestedthat an inhibitor of embryonic development was present in BOF,
and its unproductive or detri-mental offect on development of 1-cell mouse embryos decreased
when BOF dilnteel in TCM-199to the conentration of 1mg/ml.
