摘要
目的探讨转化生长因子β1(TGF-β1)在支气管哮喘发病中的作用,及利用非创伤性方法研究哮喘惠者气道炎症与气道重塑。方法4%高渗盐水诱导痰液,利用ELISA法检测哮喘患者及对照者血清、诱导痰中TGF-β1的浓度,并检测各种炎症细胞的数量。结果哮喘患者诱导疾中嗜酸细胞数量高于对照组(P<0.01),中重度哮喘患者诱导痰中的TGF -β1含量明显增加(P<0.01)。痰中嗜酸细胞与TGFβ1呈明显正相关,与肺功能呈负相关。诱导痰中TGF-β1与肺功能呈负相关,即与疾病的严重程度正相关。结论用诱导痰的方法检测哮喘患者气道炎症与气道重塑是一种安全有效的方法。嗜酸细胞、中性粒细胞在哮喘发作中均有重要意义,中重度哮喘患者TGF-β1含量明显增加,且与疾病的严重程度呈正相关,说明TGF-β1在哮喘气道重塑的过程中起着重要的作用。痰中的TGF-β1可作为气道重塑指示剂。
Objective To study the role of transforming growth factor - beta 1 and the airway inflammation and airway remodeling by non - invasive methods in asthmatic patients, Methods To examine the levels of TGF - β1 in the serum and induced sputum of the asthmatic patients and normal subjects by ELISA and to detect cell number, Results Compared with normal subjects, eosinophils of sputum were significantly increased in asthmatic patients ( P 〈 0.01 ). The levels of TGF - β1 in sputum were significantly increased in the patients with moderate and severe asthma ( P 〈 0. 01 ), There was a positive correlation between eosinophil counts and TGF - β1 in the sputum and a negative correlation between eosinophil counts in the sputum and pulmonary function, The levels of TGF - β1 in sputum negatively correlated with the FEV1/FVC%, Conclusion It is safe and effective to test airway inflammation and airway remodeling by using induced sputum method, Increased number of eosinophils and neutrophils were important in asthma, Obviously elevated levels of TGF -β1 in moderate and severe asthma and a significantly positive correlation between TGF - β1 and the seriousness of asthma were also found. TGF - β1 may play an important role in the development of airway remodeling of asthma and be an indicator of airway remodeling .
