丝裂霉素磁性纳米制剂抗肿瘤的研究 被引量:5

Anti-tumor effect of mitomycin-loaded magnetic nanospheres

摘要 目的探讨生物可降解药物丝裂霉素磁性纳米球(MMC-MNS)的释药特点,及其体内外抑制乳腺癌细胞生长的活性和特征。方法采用动态透析结合紫外分光光度法测定MMC- MNS在体外中性介质中的释药特性;以乳癌细胞株MCF-7为对象,SRB法测定MMC-MNS在4种不同药物浓度下的体外抑瘤效率;将乳腺癌皮下移植瘤裸鼠模型分为4组(分别为尾静脉注射生理盐水、MMC、空白MNS以及MMC-MNS组),瘤体表面均给予相同三维立体梯度磁场作用,观察MMC-MNS靶向治疗裸鼠乳腺癌的效果。结果MMC-MNS具有良好的缓释功能;MMC-MNS对乳癌细胞MCF-7有明显的杀伤活性,呈剂量-效应关系;经MMC-MNS处理后的肿瘤生长较其他组明显减慢,瘤重抑瘤率高达54.2%。结论MMC-MNS在体外有明显的药物缓释效应,能在较长时间维持有效作用浓度,在体内外均表现出良好的抗癌杀瘤作用,具有良好的临床抗肿瘤应用前景。 Objective To investigate the release characteristic and in vitro and in vivo antitumor effect of bio-soluble magnetic nanospheres of mitomycin-loaded albumin (MMC-MNS). Methods The dynamic dialysis assay in combination with ultraviolet spectrophotometer was used for release characteristic of nanospheres in neutral medium. SRB colorimetric assay was used to evaluate antitumor effect on breast cancer cell line MCF-7 under four different drug concentrations. The therapeutic effect of MMCMNS was also intensively studied on nude mice bearing breast cancer which were grouped by what they were injected (physiological saline, MMS, naked MNS and MMC-MNS). Results MMC-MNS had a good function of extended releasE;MCF-7 cell line was sensitive to MMC-MNS and the cytotoxity was correlated with the loaded drug dose. Treatment with MMC-MNS via intravenous administration under tri-dimensional magnetic force near the tumor showed more significant anticancer activity than any other modality. Conclusion MMC-MNS had obvious function of extended release and therapeutic effect both in vitro and in vivo,which will be promising in antitumor applications.
