摘要 目的建立高效液相色谱法测定小鼠血浆中丝裂霉素C聚氰基丙烯酸正丁酯磁性纳米球浓度的方法.方法血浆样品经乙酸乙酯提取后,以乙腈∶醋酸钠缓冲液pH(15∶85)为流动相,使用LiChroCART LiChrospher RP-18(250 mm×4 mm,5 μm)色谱柱分离;流速为1.0 ml/min;检测波长为365 nm.结果血浆中丝裂霉素C测定的线性范围为0.04~1.00μg/ml,线性方程为:Y=16388X-17.17,r=0.9998.结论该方法简单实用,结果准确,适用于丝裂霉素C-聚氰基丙烯酸正丁酯磁性纳米制剂临床前药代动力学研究. Objective To establish a method by high-performance liquid chromatography (HPLC) for determining the concentration of magnetic mitomycin C-polybutylcyanoacrylate nanoparticles in mouse plasma. Methods Chromatography was performed on a LiChroCART C18 (250 mm×4 mm, 5 μm) column with the mobile phase consisting of acetonitrile-NaAC (15:85), the flow rate of 1.0 ml/min, and the detection wavelength of 365 nm. Sample extraction was carried out with ethylacetate. Results The linear range of mouse plasma mitomycin C concentration was 0.04-1.00 μg/ml, and the linear equation of Y=16 388X-17.17 (r=0.999 8) was derived. Conclusion This method is very easy to operate and suits the need of perclinical pharmacokinetic studies of mitomycin-magnetic nanoparticles and yields accurate and precise results.
