摘要
建立了检测促卵泡素(FSH)抗体的间接ELISA方法,用该法检测由FSH-BSA免疫BaLb/c小鼠所分离的5份待检血清,结果均为阳性。通过反复试验,确立间接ELISA检测FSH抗体的最佳反应条件,即酶标二抗工作浓度为1∶1 200,抗原包被浓度10μg/mL,待测血清最佳稀释度为1∶500,封闭液为1.0%的脱脂奶粉,同时设置抗原阻断试验对结果进行验证。
An indirect enzyme-linked imnunosorbent assay(ELISA) was standardized to detect the antibody against follicular-stimulating honnone(FSH). Being tested with the indirect ELISA, 5 serum samples which were acquired by the FSH-BSA immunized BaLb/cmouses were all positive, The optimum work concentration of the indirect ELISA was established,Polystyrene micro-titer plates were coated by the purified FSH at theconcentration of 10μg/ml, The working concentration of sertan samples was 1:500 and the sheep-against-mice IgG labeled by HRP was 1 : 1 200,The confining liquid was 1,0% defatted milk powder, Antigen-blockage test was designed to check the results.
