摘要 采用ELISA法和流式细胞法,测定经卵白蛋白(OVA)免疫豚鼠的球蛋白Ig和T淋巴细胞增殖率的变化规律,研究Zmu-1:DHP豚鼠对病毒易感性的免疫学基础。结果表明:①豚鼠最佳OVA免疫剂量为0.1 mg/只;②OVA刺激后,Zmu-1:DHP豚鼠产生的特异性IgG及其亚类抗体,与DHP豚鼠基本相同;③Zmu-1:DHP豚鼠经OVA刺激,T淋巴细胞的增殖率显著低于DHP豚鼠;④OVA刺激豚鼠很少产生IgE;⑤加与不加Al(OH)3免疫比较,IgG1含量变化较大。表明Zmu-1:DHP豚鼠的细胞免疫功能较低,体液免疫功能与其他豚鼠相同,初步认为T细胞免疫功能低下,可能是引起Zmu-1:DHP豚鼠对某些病毒较敏感的原因之一。豚鼠的变态性抗体IgE产量较小;Zmu-1:DHP豚鼠适合于制作感染模型、研究免疫学机制或免疫功能的动物模型。 To investigate the immunology base of Zmu-1 : DHP guinea pig sensitive to virus infection, the Ig and T lymphocytic proliferation rate were determined by ELISA and FCM means. The suitable immune dosage of OVA is 0.1mg per guinea pig, After immunization with OVA, the level of characteristic IgG, IgG1, IgG2 in Zmu-1 : DHP guinea pig serum is approximately the same as DHP guinea pig, but its T lymphocytic proliferation rate is prominently lower than that of DHP guinea pig, The content of IgE in both of guinea pig sera is very low, The variation of IgG1 content is larger between immunization groups with or without AI( OH)3, The eytoimmtme function of Zmu-1:DHP guinea pig is lower than that of DHP guinea pig and the body fluid immune function is as the same as DHP guinea pig, The lower T lymphocytic function is probably one of the causes of arousing more sensitive to some virus in Zmu-1: DHP guinea pig. Thus Zmu-1: DHP guinea pig may fit to be used as an infectious animal model, and to investigate the immtmogenie mechanism.
