摘要
目的:建立川芎挥发油中 Z-藁本内酯和川芎内酯 A 的气相色谱含量测定法。方法:采用 SE-54(50 m×0.2 mm)石英毛细管柱,以邻苯二甲酸二乙酯为内标,柱温240℃,汽化室及检测器温度均为280℃,分流比1:30,检测器 FID。结果:Z-藁本内酯、川芎内酯 A 线性范围分别为5.23~26.15 mg·mL^(-1)(r=0.9999)及3.25~16.26 mg·mL^(-1)(r=0.9987);精密度实验的 RSD(n=5)分别为0.72%和1.0%;重复性实验的 RSD(n=5)分别为0.81%和2.1%;平均回收率(n=5)分别为100.2%(RSD=2.0%)和97.29%(RSD=2.6%)。结论:该方法准确、重现性好、简便易行,可用于质量控制。
Objective:To develop a reliable method for determination of Z - ligustilide and senkyunolide A in essential oil of Ligusticum chuartxiong Hort. by GC. Method: Using SE - 54 quartz capillary column ( 50 m × 0. 2 mm) ,the content of Z - ligustilide and senkyunolide A were determined by GC with dimethyl phthalate as internal standard. The column temperature was 240 ℃ and both the inject and detector temperature were 280 ℃. The splitting ratio was 1 : 30. The FID was used as detector. Result: The linear ranges of Z - ligustilide and senkyunolide A were 5.23 -26. 15mg·mL^-1 (r =0. 9999) and 3.25 - 16. 26 mg·mL^-1 ( r =0. 9987) ,respectively. The RSD(n =5)of precision were 0. 72% and 1.0% ,respectively. The RSD(n =5) of reproducibility were 0. 81% and 2. 1% and the average recoveries were 100. 2% ( RSD = 2. 0% ) and 97.29% ( RSD = 2. 6% ), respectively. Conclusion: An accurate, simple, rapid qualitative method with good reproducibility for control of essential oil were established by GC for determination of Z- ligustilide and senkyunolide A.
