摘要
黑曲霉(Aspergillusniger)S21经波长266um,照射剂量8mj的激光处理40秒,获得一高产果胶酶突变株SM40-6。以果胶为底物培养,该突变株发酵滤液,初筛酶活力达854μ/ml;比出发菌及对照提高2倍。液体发酵工艺条件研究表明,该菌产酶最适培养基组成为甘薯与麦比1∶4,硫胺2%。最适培养条件为:pH3.5,温度30℃,培养时间84小时。
A mutant strain SM40-6 was obtained from Aspergillus niger S21 by treatment with 8 mj of laser irradiation at 266um for 40 seconds. Enzyme activity of culture filltrate of the mutant strain was 854μ/ml, which was about 2 times of its parent strain. The optimal medium consists of sweet potato and wheat bran (1: 4). and 2% ammonium sulfate. The optimal culture conditions are as follows: initial pH3. 5, temperature 30℃ and cultivation time 84 hr.
