摘要
目的探讨门静脉右支结扎后,肝细胞凋亡与增殖的变化规律。方法SD大鼠78只,随机分成假手术对照组、右侧肝动脉与门静脉分支均结扎组(HA+PV组)和单独右侧门静脉分支结扎组(PV组),观察术后3、12、24 h及3、7、14 d肝叶的大体形态,苏木精一伊红(HE)染色、电子显微镜、末端脱氧核苻酸转移酶介导的缺口末端标记法(TUNEL)和免疫组织化学法观察肝细胞凋亡和增殖,并进行定量分析。结果右侧肝动脉与门静脉分支均结扎后,肝右叶发生凝固性坏死,肝细胞坏死,早期有少量凋亡细胞。单独右侧门静脉分支结扎后,肝右叶萎缩,3 h即出现凋亡,24 h达高峰,随后出现坏死,凋亡细胞数减少。肝左叶代偿性肥大,增殖指数增加,至14 d时最高。结论大鼠肝右叶门静脉结扎后,肝右叶细胞凋亡,左叶细胞增殖,分别以24 h和14 d最为明显,可为实验提供肝细胞凋亡与增殖功能磁共振成像研究理想的动物模型。
Objective To appraise the pathological changes of liver cells after ligation of the right portal vein. Methods 78 S-D rats were randomly divided into control group, right hepatic artery and right portal vein ligated group ( HA+ PV group), right portal vein ligated group(PV group). Rats of each group were sacrificed and sampled at 3 h, 12 h, 24 h, 3 d, 7 d, 14 d after operation. The apoptosis and proliferation of hepatic cells were observed and quantificational analysis was performed by HE staining, electron microscope, TUNEL and immunohistochemical techniques. Results In HA-q-PV group, coagulation necrosis was found in right hepatic lobe, with necrosis of liver cells found in each subsections, accompanied by a few apoptotic cells in the early periods. In PV group, right hepatic lobe atrophied, apoptosis was found at 3 h, reaching peak at 24 h and declined thereafter. The left lobe became hypertrophyied. PCNA-LI increased and reached the maximum at 14 d. Conclusions Ligation of the right portal vein induces hepatocyte apoptosis in the right hepatic lobe and proliferation in the left lobe, which are more significant at 24 h, 14 d respectively. As a result, we have developed an ideal animal model for investigation by functional MRI to reveal hepatocyte apoptosis and proliferation.
