摘要
目的分析热应激蛋白70(heat stress or stress prote ins 70,Hsp70)在苯对小鼠骨髓细胞毒性中的作用,为进一步阐明Hsp70的生物学意义和苯的防治提供科学依据。方法经过原代细胞培养后,细胞分组为:染毒组,以0、2、10、50mmol/L浓度的苯染毒2 h;热应激染毒组,细胞经40℃受热1 h,37℃恢复1 h,再以0、2、10、50mmol/L浓度的苯染毒2 h。检测Hsp70表达时的光密度,使用流式细胞术观察Hsp70表达水平和小鼠骨髓细胞凋亡程度的关系。结果小鼠骨髓细胞受热应激30m in后,Hsp70轻度增加,而应激60,90m in后,Hsp70明显升高。热应激后Hsp70表达增加;热应激组细胞除0剂量染毒组外,其他剂量组细胞发生凋亡百分比均高于染毒组。热应激后Hsp70表达增加。结论Hsp70的增加可以提供一定程度的毒物耐受能力或细胞保护功能,热应激后细胞Hsp70的增加不能抑制2,10mmol/L浓度苯所致小鼠骨髓细胞凋亡的发生。
Objective This study analyzes the effects of Hsp70 on benzene cytotoxicity of stem cells Methods After primary cell culturing, cells were randomly divided into two groups, (1) common group, cells were exposed to 0, 2,10,50 mmol/L benzene for two hours respectively; (2) heat stress group, cells were heat stressed at 40℃ for 60 min and recovery at 37℃ for 1 h, then exposed to 0, 2, 10, 50mmol/L benzene for a further two - hour. Detecting Hsp70 light dencity, Hsp70 level and degree of apoptosis was determined by flow cytometry. Results it showed that Hsp70 slightly increased when cells were heat stressed for 30 min, and significantly increased tor 60 or 90 min.So the tirst part suggested that the increase of Hsp70 may provide cells the toxic tolerance or cytoprotection to a narrow concentration range of benzene exposure. The level of Hsp70 was elevated after heat stress. Besides, for 2 and 10 mmol/L benzene exposure, percentages of apoptotic cells in Hsp70 group were higher than those in regular group except for 0 mmol/L benzene exposure. Cenclusions The increase of Hsp70 may provide cells the toxic tolerance or cytoprotection to a narrow concentration range of benzene exposure. The increase of Hsp70 level after heat stress could not protect mice bone marrow stem cells from apoptosis.
