摘要
目的 探讨热应激蛋白 70 (HSP70 )在苯致小鼠骨髓干细胞毒性中的作用。方法 取小鼠骨髓干细胞 ,体外 33℃ ,5 %CO2 孵箱中孵育过夜。将细胞于 4 0℃水浴受热 0~ 90min ,于 33℃恢复 1h ,用Westernblot方法检测细胞HSP70表达水平。另取细胞分组为 :不接触热应激组 (非热应激组 ) ,以不同浓度(0 ,2 ,10 ,5 0mmol/L)的苯染毒 2h ;热应激组 ,细胞经 4 0℃受热 1h ,33℃恢复 1h ,再以不同浓度 (0 ,2 ,10 ,5 0mmol/L)苯染毒 2h。应用MTT比色试验检测细胞活性。结果 小鼠骨髓干细胞受热应激 30min后 ,HSP70轻度增加 ,而应激 6 0 ,90min后 ,HSP70明显升高 ;热应激组细胞用 2 ,10mmol/L苯染毒 2h后 ,其细胞活性较非热应激组高。而在 0 ,5 0mmol/L苯作用下其细胞活性又低于非热应激组。结论 HSP70的增加可以提供一定程度的毒物耐受能力或细胞保护功能。
Objective To investigate the role of heat stress protein 70(HSP70)in the cytotoxicity of mice bone marrow stem cells induced by benzene.Methods Bone marrow stem cells of mice were separated and cultured in 5% CO 2 incubator at 33 ℃ overnight.The cells were then heat stressed at 40 ℃ for 0~90 min, and kept at 33 ℃ for 1 h.Western blot was used to detect the level of HSP70.Other cells were taken and randomly divided into two groups:(1)Group without heat stress.Cells were exposed to 0,2,10,50 mmol/L benzene for two hours respectively.(2)Heat stress group.Cells were heat stressed at 40 ℃ for 60 min and recovered at 33 ℃ for 1 h, then exposed to 0,2,10, 50 mmol/L benzene for a further two hour.The cytotoxicity of benzene was determined with MTT test.Results HSP70 was slightly increased after cells were heat stressed for 30 min,and significantly increased for 60 and 90 min.Compared with the group without heat stress, cell activities were higher in heat stress group after exposure to 2 and 10 mmol/L benzene for 2h,but they became lower when treated with 0 and 50 mmol/L benzene for 2h.Conclusions The increase of HSP70 may provide cells with toxic tolerance or cyto protection within a narrow concentration range of benzene exposure.
