电针刺激足三里穴对内脏痛大鼠行为学及骶髓后连合核中GFAP、OX42表达的影响 被引量:8

摘要 探讨电针刺激大鼠足三里穴(ST36)对内脏痛的镇痛作用机制。向大鼠乙状结肠注射福尔马林制作内脏痛模型。将实验大鼠分成A、B、C和D组:A组为单纯内脏痛组;B组为预先给予电针刺激足三里穴,再制作内脏痛模型;C组为制作内脏痛模型后再给予电针刺激足三里穴;D组为对照组。观察大鼠痛行为表现后,应用免疫荧光组织化学方法观察内脏痛大鼠骶髓后连合核(dorsal commissural nucleus,DCN)内星形胶质细胞的标记物胶质原纤维酸性蛋白(glial fibrillary acidic protein,GFAP)和小胶质细胞的标记物OX42的表达变化。单纯内脏痛组大鼠的痛行为表现最为明显,内脏痛+针刺组大鼠的痛行为表现略减低,针刺+内脏痛组大鼠的痛行为表现明显减弱。单纯内脏痛大鼠DCN内GFAP和OX42的表达明显增强;内脏痛+针刺组大鼠DCN中GFAP和OX42的表达明显减弱;针刺+内脏痛大鼠DCN中GFAP和OX42表达减低更加显著。实验结果表明电针刺激足三里穴能明显地减弱伤害性刺激引起的内脏痛反应,这种作用可能与星形胶质细胞和小胶质细胞有关。 In order to explore the analgesia effect of electroacupuncture (EA) at Tsusanli on visceral pain and its possible mechanism, rats were divided into four groups (group A, B, C, D) and formalin was used to induce visceralgia. Group A was visceral pain one. In group B, rats were pretreated with EA at Tsusanli before visceralgia was induced. In group C, visceral pain was induced in advance, and then EA at Tsusanli was carried out. Group D was control group. Pain behavior was observed. By using immunofluorescence histochemistry, expressions of glial fibrillary acidic protein (GFAP) and OX42, which are makers for astrocyte and microglia, respectively, in dorsal commissural nucleus (DCN) was detected. The results showed that the pain behavior of group A was the most obvious, while it decreased in group C. In group B, pain behavior decreased markedly. The expressions of GFAP and OX42 in DCN were most distinctive in group A, while it decreased in group C and reduced much significantly in group B. Our results demonstrated that EA at Tsusanli could significantly decrease the visceral pain induced by formalin. This analgesic effect may potentially relate with astrocyte and microglia.
