摘要
目的研究胃腺癌及癌旁正常组织中ODC与c-myc mRNA的表达情况,试图探讨ODC与c-myc在胃腺癌发生发展中的关系及作用。方法用RT-PCR方法,检测22例胃腺癌及其癌旁正常组织中ODC与c-myc mRNA的表达。通过与恒定表达的磷酸甘油醛脱氢酶(GAPDH,内参照)mRNA比较,计算出c-myc及ODC mRNA与GAPDH mRNA PCR产物的积分光密度比值(RM/G、RO/G),以RM/G、RO/G值分别反映各组织中c-myc及ODC mRNA表达量的高低。结果胃腺癌组织中RM/G、RO/G值明显高于癌旁正常组织,RM/G分别为0.785±0.106、0.670±0.103(P<0.01),RO/G分别为0.817±0.090、0.711±0.101(P<0.01)。低分化胃腺癌组织中RM/G、RO/G值显著高于高中分化的胃腺癌(P<0.05),尤以RO/G为甚(P<0.01)。癌及癌旁正常组织中c-myc与ODC mRNA的表达均有显著的相关性(p<0.01)。结论胃腺癌组织中c-myc及ODC mRNA的表达增加,且与恶性程度正相关;胃腺癌组织中c-myc与ODCmRNA的表达有密切的关系。c-myc及ODC在胃腺癌的发展中可能起一定的作用。
Objective To investigate the expressions of ODC mRNA and c-myc mRNA and their relationship in gastric adenocarcinoma,and to evaluate their roles in the pathogenesis and development of gastric adenocarcinoma. Methods Using RT-PCR method, the ODC and c-myc mRNAs expressions were detected in the cancer tissues and its adjacent normal mucosa of 22 patients with gastric adenocarcinoma. The ratio (RM/G or RO/G) of integrated optical density of ODC (or c-myc) mRNA PCR product to that of glyceraldehyde phosphate dehydrogenase(GAPDH) mRNA PCR product was calculated. The relative expression quantity of ODC or c-myc mRNA in tissues was represented by RM/G or RO/G respectively. Results The RM/G and RO/G were higher in gastric adenocarcinoma tissues than that in normal gastric mucosa adjacent to carcinoma tissues. In adenocarcinoma and normal tissues, the RM/Gs were 0. 785 ± 0. 106 and 0. 670 ± 0. 103, respectively (P〈0. 01), while the RO/Gs were 0. 817 ± 0. 090 and 0. 711 ± 0. 101, respectively (P〈0. 01). The RM/G and RO/G were higher in poorly differentiated adenocarcinorma than that in well and moderatedly differentiated adenocarcinorma. The expression of c-myc mRNA was closely related to ODC mRNA both in carcinoma tissues and in normal tissues(P〈0.01). Conclusion The expressions of ODC and c-myc mRNAs in gastric adenocarcinoma tissues are increased. Moreover,they are positive related to differentiation degrees of gastric adenocarcinorma, and correlated with each other. The ODC and c-myc may play a important role in the development of gastric adenocarcinoma.
