摘要
利用兼并性引物克隆出棉铃虫线粒体细胞色素氧化酶亚基Ⅱ基因(cox2),cox2基因编码框包含682个核苷酸,编码227个氨基酸的蛋白质;起始密码子为ATG,终止密码子仅有一个T组成。利用GenBank数据库搜索了其他12种昆虫的线粒体cox2基因序列,通过同源性比较,发现棉铃虫的cox2与鳞翅目4种蚕的cox2同源性最高,核苷酸和氨基酸序列同源性分别达到85%~88%和93.0%~94.7%。同时,根据cox2核苷酸和编码的氨基酸序列进行了13种昆虫的分子系统学分析的探讨,发现分子系统树与形态分类基本一致,但略有差异。
Mitochondrial cytochrome oxidase Subunit Ⅱ gene (cox2) of Helicoverpa armigera was cloned and sequenced. The open reading frame of cox2 gene contains 682 nucleotides, encoding a putative protein of 227 amino acids. The initial codon of mtcox2 of Helicoverpa armigera is ATG. The mtcox2 gene uses an incomplete termination codon, T, which can generate a complete termination codon, TAA, by polyadenylation of the transcripts. Compared with other cox2, the Helicoverpa armigera cox2 shows high identity with four cox2 from silkmoth, 85% ~ 88% in nucleotide sequence and 93.0% ~ 94.7% in amino acid sequence respectively. Based on the alignment of nucleotide and amino acid sequences, the phylogenetic tree of 13 insect cox2 genes is constructed. The molecular phylogenetic tree is similar with morphology tree.
