桂皮酸和全反式维甲酸及维生素C对HL-60细胞的分化研究 被引量:4

摘要 目的:探讨桂皮酸、全反式维甲酸(all transretinoicacid,ATRA)和维生素C(V C)对人急性早幼粒白血病(HL60细胞)的分化作用。方法:用2.5mmol/L桂皮酸、1μmol/LATRA和0.5mmol/LV C单独,和联合处理HL60细胞后,观察细胞形态改变,细胞增殖速度、硝基四氮唑蓝(NBT)还原反应和过氧化酶染色反应。结果:ATRA、桂皮素和联合组都能使HL60细胞形态向成熟粒细胞转变,对细胞增殖有明显抑制作用,能使细胞的NBT反应显著增强(P<0.01),过氧化酶染色为弱阳性或阴性;V C对细胞形态和增殖抑制第5天才出现,使NBT反应和染色反应改变不明显(P<0.05)。结论:桂皮酸、ATRA和三者联合对HL60细胞有很强分化诱导效应,V C的分化效应弱,但有增强分化的作用。 OBJECTIVE: To study the inducing differentiation effect of cinnamic acid, ATRA and vitamin C on HL-60 cells. METHODS: After the HL-60 cells treated by cinnamic acid (2.5 mmol/L) ,ATRT(1 μmol/L) and vitamin C (0. 5 mmol/L) alone and combination, the change of the cell morphology, the cell proliferation rate, NBT reduction and color reaction of peroxidase of the cells were observed. RESULTS: The HL-60 cells morphology transformed into more mature myeloid, the call proliferation was inhibited significantly and the former effect was powerful than the latter. NBT reduction and color reaction were risen significantly (P〈0. 01), color reaction of peroxidase of the cell was masculine slightly or feminine with cinnamic. The changes of NBT reduction and color reaction were not significant (P〉0. 05) for vitamin C. The differentiation was risen significantly for their combined action. CONCLUSIONS: Compounds of Cinnamic acid, ATRA and vitamin C had much differentiation effect on HL-60 cell. The differentiation effect of vitamin C is not obvious, but it would rise differential effect for cinnamic acid.
