摘要 目的观察新型维甲酸衍生物4-氨基-2-三氟甲基苯基依曲替酸酯(ATPA)对白血病NB4细胞株生长和分化的影响。方法 ATPA作用于NB4细胞后,采用四甲基偶氮唑蓝(MTT)法,检测细胞的生长抑制率;硝基四唑氮蓝(NBT)还原实验法分析细胞的分化指标;流式细胞术检测细胞表面分化抗原及细胞周期的变化情况。结果 ATPA呈浓度和时间依赖性抑制NB4细胞增殖;NBT阳性细胞率增加,表明ATPA具有诱导NB4细胞分化的作用;G0/G1期细胞表达量增加,而S期细胞表达量减少,表明细胞周期进程受影响,细胞阻滞在G0/G1期;细胞表面分化抗原CD11b表达增加,CD13表达减少。结论 ATPA对白血病细胞株NB4有明显的诱导分化作用。 Objective To study the effect of differentiation induced by 4-amino-2-trifluoromethyl-phenyl acitretinate(ATPA) on human leukemia NB4 cell.Methods NB4 cells were cultured in vitro and treated with ATPA of different concentration.The proliferation was studied by MTT test and growth curve of cell.Nitroblue tetrazolium(NBT) reduction test was used to analyze cell differentiation indexs.The cell cycle and the expression of the maturation specific cell surface markers were analyzed by flow cytometry(FCM).Results ATPA had powerful activity in inbibiting proliferation and inducing differentiation.After treated with ATPA,the growth of leukemic cells was inhibited in a dose-dependent and time-dependent manner.The NBT positive cell ratio increased which indicated that ATPA could induce differentiation of NB4 cells.The expression level of the maturation specific cell surface marker CD11b of NB4 cells were increased while the expression level of CD13 was decreased.The proportion of cells in G0/G1 phase increased while S phase cells decreased.Cell cycle progression was blocked in the G1 phase.Conclusion ATPA can induce differentiation of leukemia cell NB4.
