摘要
目的研究碘缺乏对大鼠仔鼠小脑c-fos和c-jun蛋白表达的影响。方法选择健康Wistar大鼠30只(雌性20只,雄性10只),将雌鼠随机分成碘缺乏组及对照组,每组10只。碘缺乏组大鼠于合笼前1周开始喂饲低碘饲料(含碘20~40ng/g),饮去离子水,直至仔鼠生后30d;对照组大鼠喂饲标准饲料(含碘145~186ng/g),饮自来水。取生后第20、30、60天仔鼠,测定体重、血清中游离三碘甲状腺原氨酸(FT3)和游离四碘甲状腺原氨酸(FT4)水平,并取小脑组织进行免疫组织化学链霉菌抗生物素蛋白-过氧化物酶(SP)染色,用图像分析方法分析大鼠仔鼠小脑皮质c-fos和c-jun蛋白表达情况。结果碘缺乏组仔鼠生后第20、30、60天时,体重均低于对照组(均P<0.01),血清T3、T4水平均低于对照组(均P<0.01),小脑组织c-fos和c-jun染色强度均低于对照组(均P<0.05),总积分光密度值均低于对照组(均P<0.05)。结论碘缺乏可降低大鼠仔鼠小脑组织c-fos和c-jun蛋白表达,影响神经系统的发育。
Objective To approach the mechanism of iodine and thyroid hormone action in brain development and to know whether congenital iodine deficiency may regulate the protein expressions of c-fos and c-jun in the cerebellum. Methods The female rats were randomly divided into two groups (10 in each): control group and iodine deficiency group. The dam rats in iodine deficient group were fed on iodine-deficient diet and deionized water, this treatment was started from one week before mated and was kept in the lactation until 30th postnatal day of newborn rats. Control group was treated with the normal diet and tap water during the whole experimental period. The body weight and thyoid hormone levels of newborn rats were tested and the protein expressions of c-fos and c-jun in cerebellum tissue were determined with SP immunohistochemical technique and micrograph analyses system on 20 th, 30 th and 60 th postnatal day (PN). Results The average of body weight and FT3 and FT4 in all the postnatal period were significantly decreased (P〈0.01) and the protein expression of c-fos and c-jun on 20PN, 30PN and 60PN was also down regulated significantly (P〈0.05) compared with the controls. Conclusion Iodine deficiency of mother rat may down regulate the protein expression of c-fos and c-jun of their offspring which may result in damage of cerebellum development,
