摘要
本文报道了用表面增强傅里叶变换拉曼光谱法测定的脱氧核糖核酸(DNA)水溶液及固态DNA的拉曼散射结果。实验表明,由于银膜底物对水溶液中DNA各基因的有效吸附,可在较低浓度下(1μm/ml)获得不受荧光干扰的拉曼光谱图,其频率和相对强度分布与所测固态DNA拉曼光谱有明显的差异。其中,水溶液状态下吸附在银膜上的DNA的核糖部分C=O及碱基腺嘌呤基团有关振动谱带均得到显著增强。对水溶液DNA的表面增强拉曼谱带的归属进行了讨论。
The Raman spectra of DNA obtained in aqueous solution and in solid state by surface enhanced Fourier transform Raman spectroscopy (SE--FT--RS) are presented. The experimental results show that Raman spectrum of DNA in aqueous solution without any fluorescence interference can be obtained at low concentration (1μg/ml) via adsorption groups of DNA on surface of Ag film. It is obvious that different patterns on shift and relative intensity distribution of DNA exist in aqueous solution and solid state. In aqueous solution f the Raman spectral bands of nucleic ribose C ̄O groups and adenine groups are generally enhanced. The assignment of DNA bands in aqueous solution is discussed.
