摘要
目的:观察利多卡因(lidocaine,L ido)抗LPS所致的大鼠肺泡Ⅱ型上皮(AT-Ⅱ)细胞凋亡作用。方法:体外培养的大鼠AT-Ⅱ细胞暴露到LPS和L ido中2 4 h后,采用流式细胞仪(FCM)及电镜检测AT-Ⅱ细胞凋亡和坏死率;乳酸脱氢酶(LDH)试剂盒检测LDH活性。结果:LPS可直接导致AT-Ⅱ细胞损伤,表现为AT-Ⅱ细胞坏死和凋亡发生率增高,LDH释放量增多。L ido能降低AT-Ⅱ细胞凋亡率。结论:L ido对LPS所致的AT-Ⅱ细胞凋亡和坏死有直接的抑制作用。
Objective To investigate the effect of lidocaine on LPS induced apoptosis of cultured adult rat alveolar Type Ⅱ (AT-Ⅱ ) cells. Methods Cultured cells were exposed to LPS and lidocaine for 24 hours. Apoptosis and necrosis rates of cells were detected by flow cytometry and electron microscope. The activity of lactic dehydrogenase ( LDH ) was analyzed by using LDH kits. Results LPS induced the AT-II cell injuries by increasing not only the necrosis and apoptosis rates but also the LDH release of cultured AT- Ⅱ in vitro. Lidocaine decreased the necrosis and apoptosis rates of AT- Ⅱ cells. Conclusion Lidocaine can directly inhibit the apoptosis and necrosis induced by LPS in cultured AT-Ⅱ cells.
