摘要
目的探讨阿托伐他汀对心肌肥厚时缝隙连接重构的影响。方法雄性SD大鼠随机分为3组:假手术组(SO);腹主动脉缩窄组(AC);腹主动脉缩窄+阿托伐他汀组(AA)。腹主动脉缩窄术建立肥厚模型。应用超声心动图测量心肌肥厚及采用免疫组织化学方法及免疫印迹法观察心室肌连接蛋白43(Cx43)的变化。结果AC组大鼠心室肌的Cx43排列较SO组的紊乱,AA组心室的Cx43排列较AC组的明显改善,3组之间的Cx43含量相比较无统计学差异(P>0.05)。结论阿托伐他汀能部分防止压力负荷诱导的心肌肥厚时的Cx43重构。
[Objective] To investigate the effect of atorvastatin on gap junction remodeling in hypertrophied myocardium induced by pressure-overload. [Methods] Twenty four male SD rata were divided into three groups randomly: sham operation (SO) group; abdominal aortic constrietion(AC) group; abdominal aortic constriction + atorvastatin(AA) group. We used abdominal aortic constriction operation to make cardiac hypertrophy model. Left ventrieular hypertrophy was measured with eehoeardiography. The change of eonnexin43 was observed with immunohistochemistry and Western blotting techniques. [Results] In contrast to SO group, which showed eonnexin43 distributed regularly, ventrieular myoeardium from AC group showed that the distribution of eonnexin43 was marked disorganized. Compared with AC group, the disorganization of connexin43 was improved in AA group. There was no statistical difference in the amount of connexin43 among of the three groups (P 〉0.05). [Conclusion] Atorvastatln can prevent partly gap junction remodeling of hypertrophied myoeardium induced by pressure overload.
