摘要
目的 探讨survivin、bcl-2和Ki-67在星形胶质细胞反应性增生与低级别星形胶质细胞瘤中表达的区别及意义。方法 利用组织芯片和免疫组化PV6000通用型二步法检测正常脑组织、星形胶质细胞反应性增生、低级别(Ⅰ~Ⅱ级)和高级别(Ⅲ~Ⅳ级)星形胶质细胞瘤中survivin、bcl-2和Ki-67蛋白的表达情况。结果 正常脑组织中三者均为阴性表达;survivin、bcl-2和Ki-67在星形胶质细胞反应性增生组中阳性率分别为26.7%、26.7%和22.2%;在低级别星形胶质细胞瘤组中阳性率分别为53.2%、50.0%和70.2%;在高级别星形胶质细胞瘤组中阳性率分别为88.1%、79.2%和95.2%。survivin、bcl-2和Ki-67在增生组与低级别组之间差异均具有显著性(P〈0.01)。结论 survivin和bcl-2在星形胶质细胞反应性增生及星形胶质细胞瘤的发生、发展中起重要作用;survivin、bcl-2和Ki-67联合应用在两者的鉴别诊断中可能有一定的意义。
Purpose To study the difference of survivin, bcl-2 and Ki-67 protein expression in reactive and neoplastic astrocytes and their significance. Methods Immunohistochemistry and tissue microarray techniques were used to determinate the expression of survivin, bcl-2 and Ki-67 proteins in 49 cases of astrocytes proliferation, 49 cases of low-grade astrocytomas, and 50 cases of high-grade astrocytomas. 12 cases of normal brain tissues were used as controls. Results The expression of survivin, bcl-2 and Ki-67 proteins were all negative in control group; the positive rates of expression of them in reactive astrocytes were 26.7%, 26. 7%, and 22. 2% ; in low-grade astrocytomas were 53. 2%, 50. 0% and 70. 2% ; in high-grade astrocytomas were 88. 1%, 79. 2% and 95. 2% respectively. There were all significant difference between astrocytes proliferation and low-grade astrocytomas group (P 〈 0. 01 ). Conclusions Survivin and bcl-2 protein play an important role in astrocytes proliferation and progression of astrocytomas; survivin, bcl-2 and Ki-67 proteins might be coniunctly utilized to discriminate between reactive astrocytes proliferation and low-grade astrocytomas.
