应用组织芯片技术研究COX-2、c-erbB-2和Ki-67在星形胶质细胞增生和星形胶质细胞瘤中的表达及意义 被引量:1

Expression of COX-2,c-erbB-2 and Ki-67 in astrocytic proliferation and astrocytomas: an immunohistochemical study using tissue microarray techniques

摘要 目的研究COX_2c、_erbB_2、Ki_67在星形胶质细胞增生及星形胶质细胞瘤中的表达,并探讨三者在鉴别反应性与肿瘤性星形胶质细胞方面的作用。方法利用组织芯片技术及PV6000通用型二步法免疫组化方法检测COX_2c、_erbB_2、Ki_67在正常脑组织、星形胶质细胞增生、低/高级别星形胶质细胞瘤中的表达。结果正常组织中COX_2c、_erbB_2、Ki_67表达均为(-);增生组中阳性表达率分别为28.9%、37.8%和22.2%,与正常组比较,COX_2、Ki_67表达差异不显著,c_erbB_2差异显著(P<0.05);低级别肿瘤组中三者阳性表达率分别为68.1%、63.8%和70.2%,与增生组比较,均差异显著(P<0.05);高级别肿瘤组中三者阳性表达率分别为88.1%8、3.3%和95.2%,与低级别肿瘤组比较,均差异显著(P<0.05)。COX_2c、_erbB_2与组织学分级密切相关(r值分别为0.989和0.988)。结论COX_2c、_erbB_2、Ki_67有可能成为鉴别星形胶质细胞增生与低级别星形胶质细胞瘤的客观指标,且COX_2、c_erbB_2在星形胶质细胞瘤的发生、发展过程中有重要作用。 Objective To detect the expression of COX-2, c-erbB-2 and Ki-67 in astroeytie proliferation and astroeytomas and to explore the roles of them in discrimination between reactive and neoplastic astroeytes. Methods Four tissue chips were constructed to contain 148 formalin-fixed and paraffim-embedded tissue samples including 12 cases of normal brain tissues, 49 cases of astroeytes proliferation, 49 eases of low-grade astrocytomas and 50 cases of high-grade astrocytomas, and 12 cases of normal brain tissues were used as controls markers, including COX-2, c-erbB-2 and Ki-67 were detected on the tissue array sections by immunohistoebemieal staining methods. Retsults The expression of COX-2, c-erbB-2 and Ki-67 was all negative in normal brain tissues; The positive rates of expression of them in astrocytie proliferation were 28.9%, 37.8% and 22.2% respectively, in which there was a significant difference in the expression of Ki-67 ( P〈0.05), but not COX-2 and c-erbB-2 (P〉0.05), as compared to the control group, and those in low-grade astrocytomas were 68.1%, 63.8% and 70.2% respectively, in which there was a significant difference from those in the astrocytic proliferation group. Expression of the three markers in high-grade astroeytomas were 88.1%, 83.3% and 95.2% respectively, in which there was a significant differenee in the expression of COX-2, c-erbB-2 and Ki-67 (P〈0.05), as compared to the low-grade group. The expression of COX-2 and c-erbB-2 had close correlation to histological grade. Conclusions COX-2, c-erbB-2 and Ki-67 might be objective markers to discriminate astroeytie proliferation from low-grade astroeytomas. COX-2 and c-erbB-2 may play an important role in the progression of astroeytomas.
