高脂饮食诱导胰岛素抵抗大鼠模型游离脂肪酸和胰岛素敏感性的变化 被引量:6

Changes of free fatty acid and insulin sensitivity in rat models of insulin resistance induced by high fat diet

摘要 目的:观察经高脂饮食诱导胰岛素抵抗大鼠模型的胰岛素敏感性及血清游离脂肪酸的变化情况。方法:实验于2002-04/08在重庆医科大学动物中心及附属第一医院内分泌实验室完成。选择6周龄Wistar大鼠24只,随机分为2组,即正常饲养组和高脂饲养组,每组12只。正常饲养组及高脂饲养组饲料的热量分别为13.53,18.71kJ/g。喂养6周后,应用葡萄糖钳夹技术评价胰岛素抵抗大鼠模型(葡萄糖输注率60~120越低,胰岛素抵抗越严重),血清游离脂肪酸水平应用生化比色法测定,血清三酰甘油、总胆固醇、低、高密度脂蛋白胆固醇水平应用生化酶法测定。比较两组大鼠体质量、葡萄糖输注率以及血清游离脂肪酸等的变化情况。结果:实验过程中两组各死亡2只,进入结果分析两组各10只大鼠。①两组大鼠喂养6周后血脂、游离脂肪酸水平比较:高脂饲养组大鼠的三酰甘油、游离脂肪酸水平显著高于正常饲养组[(1.24±0.34,0.61±0.12),(0.43±0.13,0.24±0.10)mmol/L(t=2.84,P<0.05;t=3.36,P<0.01)]。②两组大鼠喂养6周后体质量、血糖、胰岛素水平及葡萄糖输注率60~120比较:高脂饲养组大鼠体质量、血浆胰岛素水平显著高于正常饲养组(t=3.49,4.25,P<0.01),葡萄糖输注率60~120显著低于正常饲养组[(9.89±3.28,28.03±8.02)mg/(kg·min)](t=5.91,P<0.01)。结论:高脂饲养大鼠的葡萄糖输注率明显降低,提示高脂饮食可导致机体产生严重胰岛素抵抗,是建立胰岛素抵抗大鼠模型的有效手段。该模型同时伴有脂代谢异常,其胰岛素抵抗的形成可能与游离脂肪酸水平升高有关。 AIM: To observe changes of serum free fatty acid (FFA) and insulin sensitivity in rat mdtlels with insulin resistance (IR) induced by high fat diet. METHODS: The experiment was conducted in the Laboratory of Endocrinology, First Affiliated Hospital and Animal Center of Chongqing Medical University from April to August 2002. 24 male Wistar rats of 6 weeks old were randomized into two groups, normal feeding group and high fat feeding group with 12 rats in each group. The heat quality of feedstuff in normal feeding group and high fat group were 13.53 and 18.71 kJ/g respectively. 6 weeks after feeding, rat models with IR were evaluated with glucose ring clamp technique (the lower the glucose infusion rate 60-120:the severer the IR), level of serum FFA was assayed with chromatometry, levels of serum triglyceride (TG), total cholesterol and high/low density lipeprotein cholesterol (TC) were measured by using biochemical enzymic technique. Changes of body mass, glucose infusion rate and serum FFA in rats were compared between the two groups. RESULTS: Two rats of each group died in the experiment, and 10 rats of each group entered the final analysis. ① Comparison of blood lipid and FFA after 6 weeks feeding: Levels of TG and FFA in the high fat feeding group were significantly higher than those in the normal feeding group[( 1.24±0.34,0.61 ±0.12 ), (0.43±0.13,0.24 ±0.10) mmol/L (t =2.84, P 〈 0.05;t=3.36, P 〈 0.01 )]. ② Comparion of body mass, blood lipid, level of insulin and glucose infusion rate 60-120: Levels of body mass and serum insulin in rats of high fat feeding group were significantly higher than those in the normal feeding group (t=3.49,4.25, P 〈 0.01 ), and the glucose infusion rate 60-120 was obviously lower than that in the normal feeding group[(9.89±3.28,28.03±8.02) mg/(kg.min)](t=5.91 ,P 〈 0.01 ).
