米非司酮对不同种类人卵巢癌细胞株细胞体外作用的研究 被引量:5

摘要 目的:研究米非司酮对不同种类人卵巢上皮癌细胞株细胞体外生长的作用。方法:应用四甲基偶氮唑蓝(MTT)比色法测定不同浓度米非司酮作用不同时间对人卵巢上皮癌细胞株3AO、OVCAR3和SKOV3细胞生长的抑制作用,以流式细胞间接荧光技术检测肿瘤细胞ER、PR蛋白表达率。结果:米非司酮以剂量和时间依赖方式抑制3AO和OVCAR3细胞体外增殖活性(均P<0.05),而对SKOV3细胞生长增殖无显著影响(P>0.05)。米非司酮和地塞米松联合应用与相同浓度的米非司酮单独应用比较,3AO和OVCAR3细胞增殖抑制率无统计学意义(P>0.05)。米非司酮浓度为10μmol/L,培养48小时,3AO细胞PR表达率为12.7±1.4%,明显低于对照组44.9±1.4%(P<0.05);OVCAR3细胞PR表达率为23.5±2.1%,明显低于对照组53.4±1.9%(P<0.05);3AO、OVCAR3和SKOV3细胞ER表达率与对照组比较,均无统计学意义(均P>0.05)。结论:米非司酮以剂量和时间依赖方式抑制PR表达阳性人卵巢上皮癌3AO、OVCAR3细胞体外增殖活性,其抑制作用与PR变化相关。 Objective: To investigate the effect of Mifeprtstone to different types of human ovarian epithelial cancer cell lines in vitro. Methods: Methyl thiazolyl tetrazolium (MTT) was used to observe the growth inhibition rates of human ovarian cancer cell lines 3AO, OVCAR3 and SKOV3 by various concentrations of Mifepdstone at different times. Flow cytometn/(FCM) was used to measure the expression rotes of ER and PR protein. Results: Mifepristone inhibited the growth of 3AO and OVCAR3 cell lines effectively, depending on the action time and concentration of the medicine in vitro ( P 〈 0.05), but no effect to the cell growth of SKOV3 ( P 〉 0.05). Comparisons between Mifepristone plus Dexamethasone and Mifepristone alone, the growth inhibition rates of 3AO and OVCAR3 cell lines had no significant difference (P〉0.05) .The expression rates of PR protein of 3AO and OVCAR3 cell, which cultured with Mifepristone of 10μmol/L for 48 hours was 12.7±1.4% and 23.5 ±2.0%, respectively, which obviously lower than that of the control group (44.9±1.4% and 52.1 ±2.4%)(P〈0.05). The expressive rates of ER protein of 3AO, OVCAR3 and SKOV3 cell lines had no significant difference between Miferistone treated group and control group ( P〉 0.05). Conclusions: Mifepristone inhibited significantly the proliferation activity of PR positive 3AO and OVCAR3 ovarian carcer cell lines in a dose-time dependent manner in vitro, the inhibitive effect was related to the PR change.
