The relationship between the expression of bFGF and the activity of cell proliferation in meningiomas

摘要 目的:研究碱性成纤维细胞生长因子(bFGF)在脑膜瘤中的表达及其与细胞增殖活性间的关系,探讨bFGF在脑膜瘤发生和发展中的作用。方法:用免疫组化LSAB法检测38例脑膜瘤组织中bFGF与增殖细胞核抗原(PCNA)的表达,分析在bFGF不同病理级别脑膜瘤中的表达及其与PCNA标记指数(PCNA-L I)的关系。结果:本组38例中有22例bFGF染色阳性,阳性率为86.84%。在良性组29例中,有24例检测到bFGF的表达,阳性率为82.76%;非典型组和恶性组均有表达。与良性组比较,非典型组和恶性组的bFGF表达强度高,其差异具有显著性。PCNA-L I在良性组、非典型组和恶性组的标记指数分别为13.71±6.1%,31.72±10.7%,47.87±14.0%。非典型型组和恶性组PCNA-L I均高于良性组。bFGF的表达强度与PCNA-L I相关,随着bFGF表达强度的升高,PCNA-L I随之增加。结论:bFGF自分泌环在脑膜瘤的发生和发展过程中起重要作用,bFGF的表达与脑膜瘤细胞增殖活性有关。 Objective: To investigate the expression of bFGF and its relation to the activity of cell proliferation in meningiomas and to explore the role of bFGF in the development of meningiomas. Methods: LSAB immunohistochemical assay was used to study the expression of bFGF and PCNA in 38 cases of meningioma, The levels of bFGF expression and labeling indexes of PCNA in histopathologically different grades and their relationship were analyzed. Results: High levels of bFGF expression were detected in 33 of 38 human meningiomas, the positive rate was 86.84%. In benign group, 24 of 29 were detected, the positive rate of bFGF expression was 73. 68%%. The bFGF positive staining cells were detected in all atypical and malignant meningiomas. The labeling indexes of PCNA were 13. 71±6. 1%, 31.72±10.7 %, 47.87±14.0% respectively in these three different grades of meningiomas. The levels of bFGF expression and labeling index of PCNA were significantly higher in atypical and malignant meningiomas than those in benign types. The expression of bFGF was significantly enh;,nced with high level of PCNA. Conclusion: The autocrine loop of bFGF may play a critical role in the pathogenesis of meningioma. The overexpression of bFGF is related to the activity of cell proliferation in meningiomas. Quantitative bFGF labeling can provide meaningful pathological information.
