妊娠早期子宫NK细胞中HIF和VEGF的表达 被引量:1

Expression of HIF,VEGFC and VEGFE in uterine NK cells in early pregnancy

摘要 目的:探讨妊娠子宫自然杀伤细胞(uterinenaturalkillercell,uNK)和外周血NK细胞(peripheralnaturalkillercell,pNK)中乏氧诱导因子(hypoxiainduciblefactor,HIF)和血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)C、VEGFE的表达。方法:采用免疫磁珠法分离纯化妊娠早期uNK和pNK细胞,提取其总RNA,采用RT-PCR技术检测HIF和VEGF的mRNA在人妊娠早期uNK细胞和pNK细胞中的表达。结果:①人妊娠早期uNK细胞中HIFmRNA表达强于pNK细胞(2.31±0.19和0.12±0.02);②VEGFC和VEGFE在两种细胞中均有表达,但VEGFE在uNK中的表达强于pNK(0.82±0.03和0.10±0.01),VEGFC在两种细胞中的表达相近。结论:人妊娠早期uNK细胞HIF和VEGFE的强表达可能与uNK细胞在胎盘血管形成中的作用密切相关。 Objective: To investigate the expression of HIF, VEGFC and VEGFE in the uterine and peripheral NK ceils in early pregnancy. Methods: NK ceils in deciduas and peripheral blood was purified by mircobeads, and the total RNA was gained. The expression of HIF, VEGFC and VEGFE mRNA in uterine NK (uNK) and peripheral NK (pNK) cells was determined by semi-quantitative RT-PCR. Results: ①Excluding the expression of HIFmRNA found in uNK and pNK cells, there was a significant difference in transcription of HIF mRNA between uNK and pNK cells (2.31±0.19 and 0.12±0.02); ②Both VEGFC and VEGFE mRNA were detected in uNK and pNK cells,but the expression of VEGFE mRNA was higher in uNK than in pNK, and the expression of VEGFC was similar between uNK and pNK. Conclusion: The strong expression of HIF and VEGFE in uNK may play important roles in the vascular development of placenta in normal pregnancy.
