摘要
硫酸基含量对多糖的生物活性起着至关重要的作用.运用硫酸钡比浊法测定梅花鹿鹿茸多糖(Pi-lose Antler Polysaccarides,PAPS)的硫酸基含量.采用可见分光光度法,最大吸收波长为360 nm,测得硫酸基含量5.8%.稳定性试验表明,硫酸基在60~120μg线性关系良好(r=0.997 19),同一样品在10~40 min内检测值比较稳定.回收率为97.25%.该方法比较准确、快速、灵敏度较高,适用于动物多糖硫酸基含量的测定.
The content of sulfated group in polysaccharides functions importantly on their biological activities. Barium sulfate turbidity was used for determining the SO4^2-content in the pilose anther polysaccarides(PAPS). SO4^2-content was determined to be 5.8% on a spectrophotometer at 360 nm. In the stability test, when the SO4^2- content ranges within of 60~120μg, the linearity was good (r= 0. 997 19); and the results obtained from the same sample was stable within a time range of 10~40 min. The recovery by this method was 97.25%. The method has been proved to be reliable,rapid and sensitive. It is suitable for the determination of the SO4^2- content in animal polysaccharides.
