牙胚细胞与PLGA支架体内复合培养的实验研究

摘要 目的摸索胎鼠牙胚细胞体外培养所需的条件及细胞-支架复合物在裸鼠体内培养的条件。方法取17日龄的胎鼠第一磨牙牙胚组织消化成细胞悬液,然后种植于PLGA支架上,再将细胞支架复合物埋植于裸鼠皮下,术后40d后取出细胞支架复合物,观察细胞与支架的吸附及生长,繁殖情况。结果细胞呈单层生长;细胞为多角形,紧密镶嵌成铺路石状;牙胚细胞染成蓝色,支架组织染成红色,牙胚细胞较散在分布,细胞与支架紧密贴附,适度伸展,并有基质分泌。结论牙胚细胞支架复合物在裸鼠体内能够成活,细胞与支架紧密贴附,生长正常并有基质分泌,表明PLGA是牙组织工程良好的支架材料。 Objective To explore the external and internal condition that the rat tooth germ cells (TGCs) culture need and to explore its biocompatibility with biodegradable polymer scaffold (PLGA). Methods Take out the first molar tooth germs carefully from 17 days fetal SD rats and mince into tiny pieces. Tooth bud tissues were digested with trypsinase into single cell suspension, seed the cells onto PLGA scaffold, then implant the compounds into the subcutaneous sacculations of athymic mice. After 40 days, we took out the compounds, observe the cells' adhesion and growth peculiarity. Results Cells growth is in monolayer. The morphologic of these cells is polygonal - shaped, long spindle - shaped, large and flat spindle - shaped. The multilateral - shaped cells grow tightly in typical cobblestone appearance. We can see TGCs cling to the PLGA scaffolds tightly, proliferate and secret medium. Conclusions TGCs could live and proliferate after seeded onto PLGA scaffolds in vivo, demonstrate that the scaffolds has compatibility with cells, it is a better outer cell medium.
