摘要
目的:体外构建不同颜色的含黑素细胞表皮,检验其作为研究色素形成和光防护新模型的可行性。方法:把角质形成细胞和黑素细胞混合培养,消化后和胶原凝胶复合,接种于已经凝固的胶原凝胶支架上,置于37℃,凝胶凝固后形成一个含黑素细胞的表皮替代物。两天后移至气液面培养,观察所制表皮的颜色变化。多巴染色检测该表皮替代物结构和黑素细胞定位。用UVA照射所制表皮,观察有无颜色改变。结果:含黑素细胞的组织工程表皮在气液面上培养一周左右出现黄褐色或棕褐色,细胞浓度越高,颜色越深。构建的表皮类似正常表皮呈三维结构,在基底部可见散在分布的黑素细胞,多巴染色阳性;有的角质形成细胞核上方有黑素帽结构。构建的表皮经UVA照射后可出现色素沉着。结论:已经成功构建含黑素细胞的表皮替代物,随细胞数量和培养时间能呈现出不同深浅的黄褐色,可以作为研究色素形成和光防护的体外模型。
Objective To construct human epidermal equivalents with melanocytes and test the feasibility of being a new model to study pigmentation and photoprotection. Methods Co-cultured keratinocytes and melanocytes were re-suspended with collagens gel and laid on coagulated collagens gel support,then several epidermal equivalents with melanocytes were constructed.Two days later,we cultured the epidermal equivalents on air-fluid levels and observed their color changes.Structure of the reconstructed epidermis and the location of melanocytes were detected by Dopa dyeing.The epidermal equivalent was observed whether it could appear color change when radiated by UVA. Results The epidermal equivalents show different degree of tan obviously after being organotypic cultured for a week.It construed of several keratinocytes layer in the epidermal equivalents.The cop-like structure was found in some keratinocytes.Melanocytes could be detected in the basal layer by Dopa dyeing.The epidermal equivalent showed pigmentation after been radiated by UVA. Conclusion The epidermal equivalents containing melanocytes had been successfully constructed.They showed many shades of tan and might altered when radiated by UVA,which suggested they could be used as a new model to study pigmentation and photoprotection.
