摘要
目的 建立动脉内皮细胞 (ECs)的体外研究模型。方法 用消化法结合机械法的方法分离胎牛主动脉 ECs,在p H7.0的 M199培养液中培养。结果 用 因子相关抗原免疫荧光法以及倒置显微镜下细胞融合后呈单层生长、鹅卵石状排列 ,证实所培养的细胞为 ECs。在无内皮细胞生长因子的条件下体外连续培养的细胞达 18代。结论 在人动脉取材极为不易的情况下 ,胎牛主动脉
Objective Establishing the research model of arterial endothelial cells in vitro. Methods By means of digesting and scraping, fetal bovine arterial endothelial cells were isolated and cultured in M199 (pH7.0).Results Vascular endothelial cells can be identified by the “cobblestone” on morphology exhibited by confluent monolayers and positive staining for Ⅷ R∶Ag. In the absence of vascular endothelial cell growth factor, ECs were subcultured to the 18th generation at most.Conclusion Now that human arteries are not readily available, the culture of fetal bovine arterial endothelial cells is a good replacement.
