摘要
目的探讨糖尿病肾病(DN)时肾小管上皮细胞转化以及肝细胞生长因子(HGF)、Smad7蛋白在其中可能发挥的作用。方法60只Wistar大鼠均行右肾切除术,伤口愈合后随机分为右肾切除对照组和糖尿病组。腹腔注射链脲佐菌素诱发大鼠糖尿病模型。采用免疫组化法检测角蛋白18(CK18)、α平滑肌肌动蛋白(αSMA)、HGF和Smad7蛋白的表达;流式细胞术检测αSMA和HGF的表达;蛋白质免疫印迹法检测Smad7蛋白的表达。结果糖尿病组较对照组CK18的表达降低,αSMA的表达上升,HGF和Smad7蛋白的表达表现为先上升后下降。结论糖尿病时,肾小管上皮细胞可发生表型转化,转化为肌成纤维细胞。HGF和Smad7蛋白表达下降可能与该模型中肾小管上皮细胞转化有关。
Objective To investigate the relationship of tubular epithelial -myofibroblast transdifferentiation and the expressions of hepatocyte growth factor (HGF) and Smad7, and to elucidate the role of HGF and Smad7 in diabetic nephropathy. Methods Diabetes was induced in male Wistar rats with right nephrectomy and streptozotocin (STZ) administration. The expressions of cytokeratin 18 (CK18), α-smooth muscle actin (α- SMA), HGF and Smad7 were assayed with immunohistochemistry. The expressions of α-SMA, HGF were assayed with flow cytometry. The expression of Smad7 was assessed by Western blot. Results Compared with those in kidneys of the control group, diabetic kidneys showed down -regulated expression of CK18 and up -regulated expression of α-SMA. The expressions of HGF and Smad7 were increased significantly in the kidneys of diabetic rats at first, then they decreased gradually, but were still higher than those of control. Conclusion The tubular epithelial ceils may undergo phenotypic alterations and change to myofibroblasts. The absence of up -regulation of HGF and Smad7 may be related with the transdifferentiation of tubular ceils in this animal model.
