摘要
实验研究观察尼群地平对于自发性高血压大鼠心肌纤维化的作用。结果表明：二氢吡啶类钙拮抗剂尼群地平３０ｍｇ／ｋｇ／ｄ治疗１４周龄自发性高血压大鼠１２周，不但可以有效地降压，逆转左室肥厚，还能预防左、右心室心肌纤维化的发生。
Aim:Todeterminetheefectsofnitrendipineonbloodpressure,leftventricularhy-pertrophy(LVH)andmyocardialfibrosisofbothventriclesofspontaneouslyhypertensiverats(SHR).Method:Antihypertensivetreatmentwithcalciumchannelblockernitrendipine(30mg·kg-1/d)wasgivento14-weekoldmaleSHRs(n=8)for12weekswithagematchedSHRsandWis-tar-Kyotorats(WKY)(n=8)receivednotreatmentascontrols.Systolicbloodpressurewasmea-suredbytail-cufmethod.LVHwasdefinedaccordingtotheratiooflefttorightventricularweightsortobodyweights.Myocardialfibrosiswasevaluatedbycolagencontentandconcentrationmg/mgdrywt.inbothventricles.Hydroxyprolincontentwasmeasuredspectrophotometricalybyitsreac-tionwithEhrich′sreagent(Stegemannmethod).Andcolagencontentwascalculatebymultiplyingthehydroxyprolincontentwiththeconstant8.2.Results:nitrendipinesignificantlydecreasedsys-tolicbloodpressureinSHRs(P<0.05)althoughthevalueremainedsignificantlyhigherthanthatofWKYcontrols.Theratioofleftventricularweighttobodyweightwassignificantlydecreasedinni-trendipinetreatedcomparedwithuntreatedSHRs(P<0.05).Rightventricularweightoftheexperi-mentalgroupshowednosignificantchangeversusthatofthecontrols.Colagencontentandconcen-trationoftheleftventricleandrightventriclewerelowerinthenitrendipinetreatedgroupcomparedwiththeuntreatedgroups.Conclusions:nitrendipinecouldattenuatehypertensionanddecreasecol-lagencontentandconcentrationofbothventriclesaswelasleftventricularmassinSHRs.Soni-trendipinemighthavethepotentialtoimprovethemyocardialpathologyinessentialhypertension.
