贝那普利与螺内酯合用对大鼠肾血管性高血压的治疗作用

摘要 采用大鼠两肾一夹高血压模型,观察贝那普利(Ben)和螺内酯(Spi)合用对高血压大鼠的收缩压(SBP),左心室肥厚及左心室心肌纤维化的作用.夹肾动脉术后15周,与假手术组比,模型组SBP,左心室重(LVW)/体重(BW)和左心室胶原蛋白浓度分别增加95%,56%和78%.与模型对照组比较,Ben15mg·kg-1·d-1ig,治疗9周,SBP,LVW/BW及左心室胶原含量分别下降了30%,25%和12%;Ben15mg·kg-1·d-1ig与Spi7.5mg·kg-1·d-1ig合用9周,SBP,LVW/BW和左心室胶原浓度分别降低了42%,34%和37%,均显著低于Ben组.本研究提示。 Effects of benazepril (Ben) with spironolactone (Spi) on systolic blood pressure (SBP), left ventricular mass, myocardial fibrosis and plasma levels of renin, angiotensin Ⅱ and aldosterone were studied in renovascular hypertensive rats. In model group, at 15 weeks after two kidney one clip operation, SBP, left ventricular weight/body weight (LVW/BW) and content of left ventricular collagen were increased by 95%, 56% and 78%, respectively, as compared with sham operation group. Treated with Ben 15 mg·kg 1 ·d 1 ig for 9 weeks, SBP, LVW/BW and content of left ventricular collagen were reduced by 30%, 25% and 12%, respectively, as compared with model group. Treated with Ben 15 mg·kg 1 ·d 1 +Spi 7.5 mg·kg 1 ·d 1 ig for 9 weeks, SBP, LVW/BW and content of left ventricular collagen were reduced by 42%, 34% and 37% as compared with model, which were significantly lower than either Ben or Spi treated group. The present results suggest that the antagonizing effects on left ventricular hypertrophy and myocardial fibrosis in renovascular hypertensive rats be potentiated when Ben and Spi are used in combination.
