摘要 目的:鉴定发酵液中的产物是2-酮基-L-古龙酸(2-KLG);方法:用高效液相色谱(HPLC)法测定氧化葡萄糖酸杆菌(Gluconobacter oxydans)和蜡状芽孢杆菌(Bacillus cereus)混合发酵液中的2-KLG。采用Aminex A 27柱46×250mm;流动相为十二烷基硫酸钠(0.01mol/L)/乙腈(3%)=2/3,流速1.0mL/min,柱温30℃;用L-7420 UV检测器检测。同时还研究了流速对检测的影响。结果:该方法回收率为94.21%,RSD为4.2%。 Objective: To identified 2 - Keto - L- Gulconic Acid in the mixed broth. Alter Gluconobacter axydans and Bacillus cereus mixed fexmentation, a HPLC method is described for determination of 2- Keto- L- Gulconic Acid in the mixed culture broth.It used Aminex A27 colunto, 445 × 250mm and detected 2 - Keto- L - Gulconic Acid with L- 7420 UV detector. The column temperature was always kept at 30℃. The mobile phase was 0.01mol/L sodium dodecyl sulfate/3% acetonitrile = 2/3 at a flow rate of 1.0ml,/min. At the same fine, the relationship between the flow rate and the result of detection was studied. Results: The average callback ratio was 94.21% and RSD was 4.2%(n= 7).
