摘要
目的采用高效液相色谱法测定古龙酸固体催化转化工艺过程的多组分含量。方法色谱柱为Inertsil柱(250 mm×4.6 mm,5μm),流动相A相为乙腈、B相为0.20%TFA水溶液,梯度洗脱,流速1.0 mL/min,柱温45℃,漂移管温度105℃,雾化气体流速2.0 mL/min。结果古龙酸甲酯、维生素C钠、古龙酸质量浓度分别在0.2~3.0 g/L(r=0.998 9),0.1~2.5 g/L(r=0.999 6)和0.3~2.5 g/L(r=0.998 6)范围内与峰面积线性关系良好。结论该方法可用于指导工艺参数的优化及生产过程的质量控制。
Objective To develop a HPLC-ELSD method to separate and determine the three ingredients in the solid catalyzing and trans-forming process of gulonic acid for guiding the optimization of the technological parameters and the quality control of productive process.Methods The Inertsil chromatography column(250 mm × 4.6 mm,5 μm) was adopted.The mobile phrase: acetonitrile(A) and 0.20% TFA aqueous solution(B).The flow rate was 1.0 mL/min.The column temperature: 45 ℃ with gradient elution.The drift tube tempera-ture: 105 ℃.The flow rate of nebulizer gas was 2.0 mL/min.Results The standard curves of gulonic acid methyl ester,ascorbic acid sodium and gulonic acid were 0.2-3.0 mg/mL(r = 0.998 9),0.1-2.5 mg/mL(r = 0.999 6),0.3-2.5 mg /mL(r = 0.998 6),respectively.Conclusion The method is sensitive and accurate,which can be used to provide the essential data for technological innovation of ascorbic acid.
