利用组织芯片研究星形细胞瘤中端粒酶hTERT,p16及Topo-Ⅱa的表达及意义

Expression and significance of hTERT,p16 and Topo-Ⅱa in astrocytoma by tissue microarray

摘要 目的探讨端粒酶hTERT,p16及Topo-Ⅱa在星形细胞瘤组织中的表达及其相关性。方法应用组织芯片技术,S-P免疫组织化学方法检测64例不同级别的星形细胞瘤及10例正常脑组织中3种蛋白的表达情况。结果端粒酶hTERT,Topo-Ⅱa的蛋白表达率分别为50.0%与59.4%,随肿瘤级别的增高而增高,差异有显著性(P<0.05)。而p16在Ⅲ,Ⅳ级中蛋白表达明显低于Ⅰ,Ⅱ级星形细胞瘤及正常脑组织,且差异有显著性(P<0.05)。端粒酶hTERT蛋白表达与星形细胞瘤组织学分级、Topo-Ⅱa蛋白表达呈正相关。结论hTERT,p16及Topo-Ⅱa蛋白表达失调参与了星形细胞瘤的发生、发展,并影响星形细胞瘤的生物学行为。 Objectives To investigate the expression of hTERT, p16 and Topo- Ⅱa in astrocytoma as well as their relationship during histogenesis and development of astrocytoma. Methods The expression of hTERT, p16 and Topo- Ⅱa in 64 astrocytoma specimens with different malignant grades and 10 normal brain tissues were studied using tissue microarray and immunohistochemistory. Results The expression rate of hTERT and Topo- Ⅱa were 50 % and 59.4 % respectively, they increased along with the increase of the malignant grade (P 〈0.05). On the contrary, the expression of p16 was lower in astrocytoma of WHO grade Ⅲ/,Ⅳ than grade Ⅰ, Ⅱ and normal brain tissues and with significant statistic difference, hTERT expression showed positive correlation with the histopathological grade of astrocytoma and the expression of Topo- Ⅱa. Conclusions hTERT, p16 and Topo- Ⅱa involved in the development of astrocytoma and influence the biological behaves of astrocytoma.
