摘要 目的探讨端粒酶hTERT、P53及Ki-67在星形细胞瘤组织中的表达与病理分级的关系。方法应用组织芯片技术,链霉菌抗生物素蛋白—过氧化物酶(SP)免疫组织化学法检测56例不同级别的星形细胞瘤及10名正常脑组织中3种蛋白的表达情况。结果端粒酶hTERT、P53及Ki-67蛋白表达的阳性率分别为45.0%(25/56)、53.6%(30/56)、64.2%(36/56),三者的表达均随肿瘤级别的增高而增高,端粒酶hTERT、P53的表达与Ki-67表达呈正相关。结论端粒酶hTERT、P53及Ki-67基因的蛋白表达与肿瘤细胞的分化程度有关,可作为星形细胞瘤恶性程度的重要标记物。 Objective To study the expression and significance of hTERT, P53 and Ki-67 in astrocytoma and their relations with histogenesis and development of astrocytoma.Methods Building a tissue microarray including 204 spots and using immunhistochemical method ,the expression of hTERT, P53 and Ki-67 was investigated in 56 astrocytoma diseases and 10 normal brain tissues.Results The positive expression rates of hTERT,P53 and Ki-67 were 45.0%(25/56),53.6%(30/56)and 64.2%(36/56)respectively. The expression of hTERT, P53 and Ki-67 positively correlated with the malignant degree of astrocytoma (P<0.05); the expression of hTERT and P53 positively correlated with the expression of Ki-67 in astrocytomas.Conclusion Overexpression of P53 and Ki-67 correlated with the degree of tumor differentiation and uncontrolled cell proliferation. It may be an important marker of astrocytoma malignancy.
