摘要
利用展示技术构建的蛋白质/多肽的突变体库,可以通过高通量进行高效地分析和筛选,因此特别适用于改造蛋白质.文中综述噬菌体、细胞表面、核糖体、mRNA等展示技术的原理和方法,以及其在酶定向进化中的应用.
With the ability of conveniently assaying and analyzing a large number of mutated proteins in a high-throughput way, display systems of libraries of mutated proteins or peptides on phage or some biological resources have been used to improve proteins and enzymes. In this article, we review recent studies on display technology, which include not only the principles and methods of phage display, cell-surface display, ribosome display and mRNA display, but also their application in aspect of directed evolution of enzymes.
