摘要
目的:探讨高脂高能量饮食诱导下肥胖易感(obesity-prone,OP)大鼠与肥胖抗性(obesity-resistance,OR)大鼠腹膜后脂肪组织中脂联素基因mRNA表达的差异。方法:18只雄性SD大鼠,以高脂高能量鼠料饲养2周后,按体重增值大小分组,增重最多6只为OP组,增重最少6只为OR组。OP、OR大鼠继续饲以高脂高能量鼠料4周后,观察各大鼠体重变化和腹膜后脂肪组织湿重,并采用RT-PCR技术检测脂肪组织中脂联素基因的mRNA表达水平。结果:①高脂高能量鼠料诱导下,OP组大鼠体重、腹膜后脂肪组织湿重均明显高于OR组(P<0.01);②OP组大鼠腹膜后脂肪组织中脂联素基因的mRNA表达水平显著低于OR组(P<0.05);③大鼠腹膜后脂肪组织湿重与脂联素基因mRNA的表达水平间存在显著的负相关关系(P<0.05)。结论:相同饮食条件下,个体发生肥胖的易感性是影响脂联素水平的重要因素。
Objective: To observe the differenl expression of adiponectin mRNA in retroperitoneal adipose tissue between obesityprone (OP) rats and obesity-resistant (OR) rats on a high-fat and high-caloric diet, Methods: After 2 weeks on a high-fat and highcaloric diet (HFHCD), 18 male Sprague-Dawley rats were separated on the basis of body weight gain into two groups. The 6 rats with the biggest body weight gain were designated as OP group and the 6 rats with the smallest body weight gain were designated as OR group, The OP and OR animals received the HFHCD for four more weeks, The body weights were recorded at the time of preparing model. The wet weights of retroperitoneal fat pad were measured when rats were killed. The levels of adiponectin mRNA expression in adipose tissue were detected by RT-PCR technique, Results: ① After six weeks on a HFHCD, the OP rats body weighs and wet weights of retroperitoneal fat pad were significantly heavier than the OR rats, ② The expressions of adiponectin mRNA in retroperitoneal adipose tissue of OP rats was significantly lower than the OR rats, ③ There were outstandingly negative correlations between wet weights of retroperitoneal fat pad and expressions of adiponectin mRNA. Conclusion: Under the same condition of the HFHCD, the different expression of adiponeetin mRNA in adipose tissue between OP and OR rats may be related to the different susceptibility to obesity.
