摘要
目的了解儿童肥胖发生机制,观察高脂高能量饮食对幼鼠脂肪组织易感基因酵母染色质沉默因子1/叉头转录因子O1(Sirt1/FOXO1)表达的影响。方法选取30只4~5周龄雄性Wistar大鼠,按体质量随机分为高脂组(20只)和对照组(10只)。高脂组予高脂高能量饲料喂养4周,将体质量位于上游前1/3(7只)大鼠判定为肥胖倾向(OP)大鼠,体质量位于下游的后1/3(7只)大鼠判定肥胖抵抗(OR)大鼠,采用反转录聚合酶链反应(RT-PCR)法测定其皮下和内脏(附睾周、肾周)白色脂肪组织中Sirt1和FOXO1基因的mRNA表达水平,采用SPSS13.0软件比较各组间差异。结果高脂高能量饲料喂养大鼠体质量分化,喂养28 d时各组间体质量均值表现为OP组〉对照组〉OR组,差异有统计学意义(P〈0.05);OP与OR组大鼠皮下与内脏各处白色脂肪组织中Sirt1 mRNA和FOXO1 mRNA表达水平差异不显著(P〉0.05),且均显著低于对照组(Pa〈0.05)。结论高脂高能量饮食对大鼠脂肪组织Sirt1和FOXO1的表达均有抑制作用,这可能是高脂饮食导致肥胖及其内分泌紊乱的内在原因之一,调节Sirt1/FOXO1通路可能是纠正与肥胖有关的脂肪细胞功能紊乱的潜在治疗靶点。
Objective To understand the mechanism of obesity developing in children and observe the effect of high fat and high energy diet on expression of susceptible genes - silent information regulator 1 ( Sift1 ) and forkhead transcription factor 1 ( FOXO1 ) in white fat tissue. Methods Thirty 4 - 5 week - old male Wistar rats were randomly divided into high fat group ( n = 20 ) and control group ( n = 10 ). After the high fat group rats were fed with high fat and high energy diet for 4 weeks. The 7 rats with the highest body weight were designated as obesity - prone (OP) rats. And the 7 rats with the lowest body weight were designated as obesity - resist(OR) rats. Reverse transcriptase - polymerase chain reaction( RT -PCR) was used to measure the expression of Sirt1 mRNA and FOXO1 mRNA in subcutaneous and visceral (epididymal and retroperitoneal) white fat tissue of young rats,then compared the expressive differences in different groups. Different groups were comparied with SPSS 13.0 software. Results Body weight differed in high fat and high energy diet fed animals, after 28 days intervention the body weight mean showed OP group 〉 control group 〉 OR group,moreover the differences were signifieant( P 〈 0. 05 ) ;there were no significant differences in the Sirt1 and FOXO1 mRNA expression between OP and OR rats in subcutaneous and visceral white fat tissue ( P 〉 0. 05 ) , however,both genes in the 2 groups' expression level significantly were lower than that in control group(P 〈 0.05 ). Conclusions High fat and high energy intake could decrease Sirt1 and FOXO1 mRNA expression in white fat tissue ,which might be one of the internal reasons that high fat diet lead to obesity, regulating Sirt1/FOXO1 way might be a potent target goal for treating dysfunction of lipocytes involved in obesity.
