宽叶缬草提取物对戊四氮致癫大鼠海马苔藓纤维发芽的影响 被引量:3

摘要 目的观察戊四氮致癫大鼠海马苔藓纤维发芽(MFS)的情况及宽叶缬草提取物的影响,探讨宽叶缬草的抗癫痫作用。方法用戊四氮37.5mg/kg腹腔注射Wistar大鼠28天建立慢性癫痫模型,同时灌胃宽叶缬草提取物500mg/kg、1000mg/kg、1500mg/kg每天3次,观察大鼠行为及脑电图变化,Timm染色观察海马苔藓纤维发芽,并用彩色病理图文分析系统分析MFS的面密度。结果大鼠28天点燃后MFS明显增加,与模型对照组比较,宽叶缬草治疗大鼠癫痫发作次数明显减少,且发作时间缩短。结论宽叶缬草提取物能对抗戊四氮的点燃作用,并抑制海马的苔藓纤维发芽。 Objective: To investigate the effect of Valeriana officinalis var. latifolia (VOL) extract on mossy fiber sprouting (MFS) in hippocampus in epileptic rats induced by pentyleneterazole (PTZ). Methods: A kindling model of chronic epilepsy in rats was established by injecting PTZ intraperitoneally. The epileptic rats in VOL groups treatment with Vol extract 3 times per day (500mg/kg, 1000mg/kg and 1500mg/ kg per time respectively). The effect of VOL on behavior and EEG in epileptic rats was examined. In addition, Timm histochemical technique was adopted to study the mossy fiber sprouting in hippocampus. Results: MFS was increased markedly in kindling rats post 28 days (0. 0812 ± 0. 0084 vs 0. 0097 ± 0. 0021 ), and which significantly reduce in VOL treatment groups(0. 0577 ± 0. 0038,0. 0314 ± 0. 0027,0. 0296 ± 0. 0033 respectively). The frequency and the duration of epileptic discharges were decreased in these treatment groups. Conclusion: VOL extract had significant antiepileptic effects on PTZ-induced epilepsy in rats, which occurred dose-dependently, and the effect might be relation with inhibiting MFS in hippocampus.
