摘要
目的:观察重组人生长激素对体外成骨细胞直接、短期的影响。方法:将不同浓度的重组人生长激素(recombinanthumangrowthhormone,RHGH)加入大鼠成骨细胞培养体系,观察不同作用时间点对大鼠成骨细胞的增殖、碱性磷酸酶活性及细胞周期的影响。结果:RHGH对成骨细胞增殖、碱性磷酸酶活性均有影响,它可促进成骨细胞增殖,提高其碱性磷酸酶活性,且可促使细胞周期中S期、G2/M期细胞百分比增加,其作用强度随浓度增加有增强趋势。结论:RHGH对成骨细胞的合成代谢有直接影响,对成骨细胞的增殖、碱性磷酸酶活性影响远强于雌激素。
AIM:To investigate the direct, short term effects of recombinant human growth hormone ( RHGH ) on rats osteoblasts cultured in vitro. METHODS :Osteoblastic cells from rats were incubated by adding rh- gh at different doses in different time,and then cell proliferation,the activity of alkaline phosphatase (ALP) and cell cycle were observed. RESULTS:RHGH stimulate proliferation and ALP activity of osteoblasts. At the same time, S phase and G2/M phase percentage was significantly higher as compared with the control. The effects tended to get stronger as the concentrations increased. CONCLUSION:The results obstained in this study suggest that rh -gh exerts direct anabolic effects on osteoblasts. The effects of rh - gh were higher than the estrogen.
